Validasi Penentuan Amonium Dalam Air Sungai Cibeureum Secara Spektrofotometri Sinar Tampak (SNI 06-2479-1991)

Amonium merupakan salah satu senyawa yang terdapat di dalam air sungai. Keberadaan amonium dengan konsentrasi 1 mg/L dapat menyebabkan kematian bagi biota air karena kekurangan oksigen terlarut. Pada penelitian ini dilakukan validasi metode penentuan amonium dalam air Sungai Cibeureum secara spektrofotometri sinar tampak. Penentuan kadar amonium dilakukan dengan metode Nessler berdasarkan SNI 06-2479-1991, yang diukur pada panjang gelombang 425 nm. Parameter pengujian meliputi linieritas (metode kurva kalibrasi), akurasi (metode kurva kalibrasi dan metode standar adisi), presisi (metode kurva kalibrasi dan metode standar adisi), batas kuantitasi (LOQ), dan batas deteksi (LOD). Hasil pengukuran terhadap parameter linieritas didapatkan nilai koefisien korelasi (r) sebesar 0,9981, persentase perolehan kembali untuk metode kurva kalibrasi dan metode standar adisi berturut-turut sebesar 96,16 % dan 94,78 %, nilai simpangan baku relatif (% RSD) untuk metode kurva kalibrasi dan metode standar adisi berturut-turut sebesar 4,48 % dan 3,27 %, nilai batas kuantitasi (LOQ) sebesar 0,04 mg/L, dan nilai batas deteksi (LOD) sebesar 0,01 mg/L. Pengukuran dengan metode kurva kalibrasi maupun metode standar adisi secara statistik tidak menunjukkan perbedaan yang signifikan pada aras 5 %. Tetapi jika ditinjau dari segi ketelitian metode standar adisi memberikan hasil yang lebih baik, sebaliknya metode kurva kalibrasi untuk konsentrasi 5,8 mg/L mempunyai ketepatan yang baik dalam penentuan amonium dalam air Sungai Cibeureum secara spektrofotometri sinar tampak. Ammonium was one of the compounds found in the river. The existence of ammonium with concentration of 1 mg/L can be caused death for aquatic biota because dissolved oxygen deficiency. In this research were performed method validation of determining the ammonium in the Sungai Cibeureum by using visible spectrophotometry. Determination of ammonium were performed with Nessler method based on SNI 06-2479-1991, measured at a wavelength of 425 nm. The test parameter include linearity (calibration curve method), accuracy (calibration curve method and standard addition method), precision (calibration curve method and standard addition method), Limit of Quantitation (LOQ) and Limit of Detection (LOD). Result of measurement of the linearity parameter obtained correlation coefficient (r) was 0.9981, the percentage of recovery for calibration curve method and standard addition method were respectively by 96.16 % and 94.78 %, relative standard deviation values (% RSD) for calibration curve method and standard addition method were respectively by 4.48 % and 3.27 %, limit of quantitation value (LOQ) was 0.04 mg/L, and limit of detection value (LOD) was 0.01 mg/L. Measurement with calibration curve method and standard addition method statistically were not shown significant differences at the level of 5 %. But if viewed in terms of precision standard addition method gave better results, whereas calibration curve method for concentration of 5.8 mg/L had a good accuracy in determination of ammonium in Sungai Cibeureum by using visible spectrophotometry.
